Toughy! Probably between Monkey Island 2, Sensible World of Soccer 97 (Amiga with a Megadrive Pad) and Baldurs Gate II.
Would be Half-Life for me, don't think theres any other game that I've played for 10 years that I still play and have fun with today. Was playing stock HLDM this past weekend and it never gets old
Favourite Game of all time Tomb Raider 1, Baldur's Gate 1 and 2, Morrowind IM MEEN, anyone ever heard of that one? Was my first PC game and got me hooked. I borrowed it from the library (it was supposed to be educational - you had to correct spelling and grammar to progress through the game). My young daughter corrected the paragraphs and I did the scary bits - killing the baddies! Still have fond memories of that!
Baldur's gate 1 & 2 C & C, Red Alert..... Morrowind, Oblivion Monkey Island 1,2,3.... M.U.L.E & Space Trader on the Commodore 64 Halo 1,2,3 ANNO Civ 1,2,3,4 Steel Battalion Silent Service Harvest Moon... There are just too many to only have one favourite.
+rep, an incredible game. For the uninitiated: Hour One: the horror of one man's miserable 80s bachelor pad. Pet rats and poor cursor recognition form the basis of the puzzles. Hour Two: the journey of terror moves into his drab office workspace. Flirtation is performed. Emails are checked. The last three. Shizzle gets real. Body horror ensues and terrible acting gets...more terrible.
Master System - Enduro Racer NES - Megaman 4? Amiga - Speedball 2, Lotus Esprit Racing (busted so many joysticks!) Megadrive - Roadrash 1 and 2 (vent that anger), Madden NFL (TOUCHDOWN!!!), NHL whatever the year (CRUNCH!!) PS1 - Time Crisis (went through so many light guns!!) PS2 - Burnout 2 and 3 (loved using aftertouch, why isn't it in Paradise?), Gran Tourismo 3 PC - Stalker SoC, FEAR
Funny, I'm not sure if its the best game I have ever played but whenever somone asks my favourite game, I always remember it quite clearly. Grim Fandango <3 this game
Nothing has captured my imagination like S.T.A.L.K.E.R. Soc...I even read "A roadside picnic" after playing it - I don't usually read books for fun!
Racing: Grid (Xbox 360) Sport: 3 Lions (PS1) FP: Battlefield 2 / Bad Company (PC/Xbox 360) RTS: C&C Tiberian Sun (PC) Point & Click: Simon the Sorcer (PC) Fighting: Mortal Kombat 2 (Megadrive) Puzzle: Lemmings (Amiga)
Final Fantasy ...the first one released in the US on the Nintendo. This was the grandaddy of the series and pretty much all other RPGs that came to be. Although my original NES died about 7 yrs ago, I still play the ROM from time to time.

To narrow it down to one or even a few would be too hard. Some lists are incomplete and some I haven't started yet, it's an ongoing project.
Planscape Torment - probably the best RPG ever. Far too many favorites from Speccy/C64 days although I still play through Last Ninja on C64.
I think some people are getting carried away and naming every game they own... There a quite a few, especially from different genres but the game I remember most fondly and had the most fun with is: Neverwinter Nights. But there are lots of Amiga/C64 games I could have listed here.
Racing: Metropolis Street Racer (Dreamcast) Sport: Fifa 98 Road to world cup. FPS: CoD4 MW1. RTS: C&C Red Alert 2 Fighting: Soul Calibur Other: Oblivion, GTA San Andreas
